用免疫组织化学PAP法,在相邻切片上分别显示生后不同时间大鼠胰岛内胰岛淀粉样多肽免疫反应(IAPP-IR)细胞和胰岛素免疫反应(Ins-IR)细胞,对胰岛内IAPP-IR和Ins-IR细胞的数密度(Nv)进行了形态计量学研究。结果表明,IAPP-IR细胞的Nv在生后1d较低,以后逐渐升高,至18d时出现高峰,以后渐降低,45d后趋于稳定。Ins-IR细胞的Nv始终较IAPP-IR细胞的Nv低,但两者在生后不同时期的变化基本呈平行关系,唯有18d时IAPP-IR细胞的Nv较Ins-IR细胞的Nv明显升高(p<0.01)。免疫组织化学法在相邻切片分别显示IAPP-IR和生长抑素免疫反应(SS-IR)细胞证明,IAPP也存在于一部分D细胞内。本研究用形态计量学方法证明了在生后发育期间IAPP与Ins在B细胞内共存,两者表达呈平行关系。同时还证明了IAPP与SS在一部分D细胞内共存。本文对上述结果的意义进行了讨论。
Immunohistochemical PAP method was used to detect the expression of IAPP-IR and Ins-IR in rat islets at different time points after birth in adjacent sections. Morphometric studies were performed on the number density (Nv) of IR and Ins-IR cells. The results showed that the Nv of IAPP-IR cells was lower at 1 day after birth and then gradually increased, reaching a peak at 18 days, then gradually decreased and then stabilized after 45 days. The Nv of Ins-IR cells was always lower than that of IAPP-IR cells, but the changes of them were basically parallel at different time points after birth. Only Nv of IAPP-IR cells was significantly higher than that of Ins-IR cells High (p <0.01). Immunohistochemistry showed that IAPP-IR and somatostatin-immunoreactive (SS-IR) cells in adjacent sections showed that IAPP was also present in some D cells. In this study, we used morphometric methods to prove that IAPP and Ins coexist in B cells during postnatal development, and the two are in a parallel relationship. It also demonstrated that IAPP and SS coexist in a subset of D cells. This article discusses the significance of the above results.
