目的:研究天钩降压胶囊中钩藤碱、异钩藤碱在大鼠肠道的吸收动力学。方法:采用大鼠在体肠灌流试验,利用HPLC测定钩藤碱、异钩藤碱的含量KROMASIL C18色谱柱(4.6 mm×200 mm,5μm),流动相乙腈-水(含0.01 mol.L-1三乙胺,冰醋酸调pH 7.5)(45∶55),流速1 mL.min-1,检测波长254 nm,柱温30℃,进样量20μL,用紫外-可见分光光度法在560nm测定酚红的含量。结果:钩藤碱吸收动力学方程为Y=-0.331 7X-0.771 8(r=0.998 5),Ka=0.331 7(h-1),t1/2=2.09(h);异钩藤碱吸收动力学方程为Y=-0.687 0X-0.150 4(r=0.985 2),Ka=0.687 0(h-1),t1/2=1.01(h)。结论:天钩降压胶囊中钩藤碱、异钩藤碱在大鼠在体小肠的吸收呈一级动力学过程。
OBJECTIVE: To study the absorption kinetics of rhynchophylline and isorhline in rat intestinal tract in Tian hookjiangya capsule. Methods: The rat intestinal perfused test was used to determine the content of rhynchophylline and isorhamine in KROMASIL C18 column (4.6 mm × 200 mm, 5 μm). The mobile phase of acetonitrile-water (containing 0.01 mol·L- 1 triethylamine and glacial acetic acid adjusted to pH 7.5), the flow rate was 1 mL.min-1, the detection wavelength was 254 nm, the column temperature was 30 ℃, and the injection volume was 20 μL. The concentration was determined by UV-Vis spectrophotometry at 560 nm Phenol red content. Results: The absorption kinetics of rhynchophylline was Y = -0.331 7X-0.771 8 (r = 0.998 5), Ka = 0.331 7 (h-1) and t1 / 2 = 2.09 The equation of the equation is Y = -0.687 0X-0.150 4 (r = 0.985 2), Ka = 0.687 0 (h-1), t1 / 2 = 1.01 (h). CONCLUSION: The absorption of rhynchophylline and isorhamine in Tianxiaojiangya Capsule in the rat intestine is a first-order kinetic process.
