目的 :构建新城疫病毒 (NDV )血凝素神经氨酸酶 (HN)基因真核表达质粒 ,并初步探索其在抗肿瘤治疗中的机制及应用价值。方法 :用 RT- PCR从 NDV中克隆 HN c DNA,并构建 HN的真核表达载体 ,转染 COS- 7细胞表达。注射至小鼠B16黑素瘤模型 ,检测其抗肿瘤作用。结果 :成功地克隆了 NDV HN c DNA,所构建的 pc DNA3- HN有良好的表达 ,动物实验显示 ,pc DNA3- HN有增强荷瘤小鼠 CTL ,NK杀伤活性的作用。结论 :NDV HN的真核表达质粒具有增强荷瘤小鼠抗肿瘤CTL ,NK细胞反应的作用。
Objective: To construct eukaryotic expression plasmid of hemagglutinin neuraminidase (HN) gene of Newcastle disease virus (NDV) and to explore its mechanism and application value in antitumor therapy. Methods: HN c DNA was cloned from NDV by RT-PCR and eukaryotic expression vector of HN was constructed and transfected into COS-7 cells for expression. Injected into mouse B16 melanoma model to detect its anti-tumor effect. Results: NDV HN c DNA was successfully cloned, and pcDNA3 - HN was expressed well. Animal experiments showed that pcDNA3 - HN could enhance CTL and NK cytotoxicity in tumor - bearing mice. Conclusion: The eukaryotic expression plasmid of NDV HN can enhance the anti-tumor CTL and NK cell responses in tumor-bearing mice.
