目的 研究表明酒石酸锑钾能抑制白血病细胞及淋巴瘤细胞生长,并诱导其凋亡。本研究旨在探讨酒石酸锑钾诱导人结肠癌SW480凋亡的作用。方法 将不同浓度酒石酸锑钾作用于体外培养人结肠癌SW480细胞。采用MTT法检测酒石酸锑钾对结肠癌SW480细胞的生长抑制作用,并计算生长抑制率;流式细胞术检测亚二倍体峰,并利用TUNEL法进行凋亡的分子生物学检测。结果 酒石酸锑钾能明显抑制结肠癌细胞生长,抑制作用呈时间和剂量依赖性;流式细胞仪检测到亚二倍体峰,亚二倍体细胞比例随时间延长而增加。TUNEL法观察到棕褐色着染的凋亡阳性细胞。结论 酒石酸锑钾能抑制SW480细胞生长并诱导细胞凋亡。
AIM: Studies have shown that antimony potassium tartrate can inhibit the growth of leukemia cells and lymphoma cells and induce apoptosis. This study aimed to investigate the anti-apoptosis effect of antimony potassium tartrate on human colon cancer SW480 cells. Methods Different concentrations of antimony potassium tartrate were used to culture human colon cancer SW480 cells in vitro. The inhibitory effect of antimony potassium tartrate on colon cancer SW480 cells was detected by MTT assay and the growth inhibition rate was calculated. The sub-diploid peak was detected by flow cytometry, and the molecular biology of apoptosis was detected by TUNEL assay. Results Antimony potassium tartrate significantly inhibited the growth of colon cancer cells in a time and dose-dependent manner. The sub-diploid peak was detected by flow cytometry and the proportion of sub-diploid cells increased with time. TUNEL method was observed in tan staining of apoptotic cells. Conclusion Antimony potassium tartrate can inhibit the growth of SW480 cells and induce apoptosis.
