目的探讨采用蛋白质组学技术研究维甲酸的耐药机制。方法提取维甲酸敏感细胞株NB4及维甲酸耐药细胞株MR2的总蛋白,进行双向凝胶电泳识别差异表达蛋白,经质谱分析、数据库搜索进行蛋白鉴定。结果单张凝胶电泳图上呈现约600多个蛋白点,组间的匹配性较好,初步筛选出三个表达差异蛋白,经鉴定分别为DJ-1蛋白、热休克蛋白70及KIAA1289未知功能蛋白。结论应用双向电泳连接质谱的蛋白质组学方法有助于筛选到耐药相关的表达蛋白,为进一步研究维甲酸耐药机制提供分子基础。
Objective To investigate the proteomic mechanism of retinoic acid by proteomics. Methods The total protein of retinoic acid sensitive cell line NB4 and the retinoic acid resistant cell line MR2 was extracted, and the differentially expressed proteins were identified by two-dimensional gel electrophoresis. The proteins were identified by mass spectrometry and database search. Results The single gel electrophoresis image showed more than 600 protein spots. The match between the two groups was good. Three differential proteins were screened and identified as DJ-1 protein, HSP70 and KIAA1289, respectively protein. Conclusion The proteomics method of two-dimensional electrophoresis coupled mass spectrometry can help to screen the expression of drug resistance-related proteins and provide the molecular basis for the further study of the resistance mechanism of retinoic acid.
