目的:探讨川芎嗪是否可通过调节血管内皮生长因子(VEGF)表达而影响大鼠实验性关节炎的发展。方法:建立大鼠佐剂性关节炎(AIA)模型,灌胃给药川芎嗪,检测大鼠足垫厚度的变化,免疫组化SP法染色CD34检测滑膜微血管密度(MVD),放射免疫法分析大鼠血清中肿瘤坏死因子(TNF-α)的含量,Western blot分析滑膜组织VEGF的表达。结果:与模型组相比较,100 mg.kg-1川芎嗪组大鼠足垫厚度显著减小,血清中TNF-α的含量和滑膜组织VEGF的表达均明显降低。结论:川芎嗪抑制大鼠佐剂性关节炎的发展与抑制病变关节滑膜VEGF的表达密切相关。
Objective: To investigate whether tetramethylpyrazine can affect the development of experimental arthritis in rats by regulating the expression of vascular endothelial growth factor (VEGF). METHODS: A rat model of adjuvant arthritis (AIA) was established by intragastric administration of tetramethylpyrazine. The thickness of rat footpad was measured by immunohistochemistry. The immunohistochemical SP method was used to detect the microvessel density (MVD) of synovial membrane and radioimmunoassay. The content of tumor necrosis factor (TNF-α) in rat serum was analyzed, and the expression of VEGF in synovial tissue was analyzed by Western blot. RESULTS: Compared with the model group, the thickness of the footpad in the rats of the 100 mg.kg-1 tetramethylpyrazine group was significantly reduced, and the content of TNF-α in the serum and the expression of VEGF in the synovial tissue were significantly reduced. Conclusion: Tetramethylpyrazine inhibits the development of adjuvant arthritis in rats and is closely related to the inhibition of VEGF expression in the synovial membrane of the affected joints.
