目的观察不同剂量双酚A(BPA)对雌性F344大鼠椎体及股骨骨密度的影响。方法 4周龄雌性F344大鼠,体质量(53.90±4.23)g,随机分为正常对照组、BPA50组[50 mg/(kg·d)]、BPA200组[200 mg/(kg·d)]、BPA400组[400 mg/(kg·d)]和苯甲酸雌二醇(EB)组[1 mg/(kg·3 d)],每组8只。双酚A(溶解于玉米油中)采用灌胃方式给药,EB行腹腔注射给药,正常对照组给予溶剂玉米油等体积灌胃。观察并记录各组精神状态、体毛脱落及体质量变化。药物干预13周后于10%水合氯醛麻醉下,利用双能X线骨密度仪测定椎体及右侧股骨骨密度。结果 BPA200组体质量最低,BPA200、BPA400组体质量与正常对照组比较均降低,差异有统计学意义(P=0.003;P=0.015)。EB组体质量最高,与正常对照组相比差异有统计学意义(P=0.014)。BPA200组椎体骨密度最低,与正常对照组相比差异有统计学意义(P=0.036)。BPA50组右侧股骨骨密度最低,较正常对照组差异有统计学意义(P=0.027)。EB组椎体骨密度较正常对照组明显增加(P=0.018)。结论一定剂量的双酚A可明显抑制大鼠体质量增长并显著降低大鼠椎体及股骨骨密度。
Objective To observe the effect of different doses of bisphenol A (BPA) on the bone density of vertebral body and femur in female F344 rats. Methods Four-week-old female F344 rats (53.90 ± 4.23g) were randomly divided into normal control group, BPA50 group [50 mg / (kg · d)] and BPA200 group [200 mg / (kg · d) , BPA400 [400 mg / (kg · d)] and estradiol benzoate (EB) [1 mg / (kg · 3 d)]. Bisphenol A (dissolved in corn oil) was given by gavage, EB by intraperitoneal injection, and normal control group by solvent corn oil and other volume gavage. Observe and record the mental status of each group, body hair loss and body weight changes. After 13 weeks of drug intervention, bone mineral density of the vertebral body and the right femur were measured by dual energy X-ray absorptiometry under anesthesia with 10% chloral hydrate. Results The body weight of BPA200 group was the lowest. The body weight of BPA200 and BPA400 groups were lower than that of the normal control group, the difference was statistically significant (P = 0.003; P = 0.015). The body weight of EB group was the highest, with significant difference compared with the normal control group (P = 0.014). BPA200 group, the lowest vertebral bone density, compared with the normal control group, the difference was statistically significant (P = 0.036). The right femur BMD of BPA50 group was the lowest, which was significantly different from the normal control group (P = 0.027). The vertebral bone density in EB group was significantly higher than that in normal control group (P = 0.018). Conclusion A certain dose of bisphenol A can significantly inhibit the growth of rats and significantly reduce the vertebral body and femur bone mineral density.
