目的探讨膜联蛋白5(annexin5)对雄性SD大鼠体重、睾丸系数、附睾系数、精子相对计数和睾酮水平的影响。方法给雄性SD大鼠腹腔注射3个不同剂量(7.5μg/kg、15μg/kg、30 μg/kg)的annexin5,对照组腹腔注射等量的pH 8.0 Tris-HCl,1次/d,连续20d。分别称重对照组和实验组SD大鼠体重、睾丸、附睾,并对附睾尾进行精子计数。HE染色观察睾丸组织结构。运用化学发光法检测对照组和各实验组SD大鼠血清中睾酮浓度。结果与对照组相比,15 μg/kg实验组大鼠附睾系数与精子相对计数均显著提高,分别提高了12.5%(131.8±9.6vs 1 17.2±5.9)(P<0.01)和31.4%(36.8±5.6vs 31.7±5.3)(P<0.05);其它剂量组与对照组相比差异均无统计学意义(P>0.05)。HE染色显示各剂量实验组与对照组相比,睾丸的形态结构没有发生明显改变。7.5 μg/kg组和15 μg/kg组大鼠血清睾酮含量显著高于对照组,分别提高了35.5%(36.33±3.89vs26.82±3.75)(P<0.01)和82.8%(49.04±5.17vs26.82±3.75)(P<0.01),30 μg/kg组大鼠睾酮含量虽也有升高,但与对照组比较,结果无统计学意义(P>0.05)。结论腹腔注射annexin5能影响大鼠睾丸生精、附睾功能及睾酮水平,并与剂量有关。
Objective To investigate the effects of annexin 5 on body weight, testis coefficient, epididymis coefficient, relative sperm count and testosterone in male SD rats. Methods Male Sprague-Dawley rats were injected intraperitoneally with 3 different doses (7.5μg / kg, 15μg / kg, 30μg / kg) of annexin 5. The control group was intraperitoneally injected with the same volume of pH 8.0 Tris-HCl once a day for 20 days . Body weight, testis and epididymis of SD rats in control group and experimental group were respectively weighed, and sperm count was performed on epididymis tail. HE staining to observe the testicular tissue structure. The concentrations of testosterone in serum of SD rats in control group and experimental groups were detected by chemiluminescence method. Results Compared with the control group, the epididymal coefficient and relative sperm count in the 15 μg / kg experimental group were significantly increased by 12.5% (131.8 ± 9.6 vs 17.2 ± 5.9) and 31.4% (36.8%, respectively) ± 5.6 vs 31.7 ± 5.3) (P <0.05). There was no significant difference in the other dose groups compared with the control group (P> 0.05). HE staining showed that the morphological structure of testis did not change significantly compared with the control group. Serum testosterone levels were significantly higher in the 7.5 μg / kg and 15 μg / kg groups than in the control group (35.5%, 36.33 ± 3.89 vs 26.82 ± 3.75, P <0.01) and 82.8% (49.04 ± 5.17 vs 26, .82 ± 3.75) (P <0.01). Compared with the control group, the testosterone level in 30 μg / kg group was also increased, but the result was not statistically significant (P> 0.05). Conclusion Intraperitoneal injection of annexin 5 can affect the spermatogenesis, epididymis function and testosterone level in testis of rats and is dose-dependent.
