目的研究Furin基因沉默对宫颈癌HeLa细胞株生长转移的影响。方法脂质体法将质粒Furin siRNA转染人宫颈癌HeLa细胞。MTT法检测细胞的增殖情况,Boyden细胞迁移和侵袭实验观察HeLa细胞的迁移和侵袭能力。Western blot检测Furin及产物MT1-MMP蛋白量的变化,ELISA法检测细胞上清中MMP2的浓度。结果细胞增殖实验中Furin siRNA处理后48h的HeLa细胞与空质粒转染组相比体外生长抑制38.6%,差异有统计学意义(P<0.01);细胞迁移和侵袭实验中,Furin siRNA处理后,HeLa细胞穿过滤膜和基质胶的细胞数量均明显少于空质粒转染(P<0.05);它可显著降低MT1-MMP蛋白表达及MMP2的分泌。结论 Furin siRNA可有效抑制人宫颈癌HeLa细胞的生长及肿瘤细胞转移,机制可能与降低Furin产物MT1-MMP的表达及MMP2合成有关。
Objective To study the effect of Furin gene silencing on the growth and metastasis of cervical cancer HeLa cell line. Methods The plasmid Furin siRNA was transfected into human cervical cancer HeLa cells by liposome method. The proliferation of HeLa cells was detected by MTT assay. The migration and invasion of HeLa cells were observed by Boyden cell migration and invasion assay. The protein level of Furin and MT1-MMP was detected by Western blot. The concentration of MMP2 in the supernatant was detected by ELISA. Results The proliferation of HeLa cells treated with Furin siRNA for 48h was 38.6% lower than that of empty plasmid transfected cells (P <0.01). In the cell migration and invasion assay, after Furin siRNA treatment, The number of cells passing through the membrane and matrigel in HeLa cells was significantly less than that in empty plasmid transfected HeLa cells (P <0.05). It could significantly reduce the MT1-MMP protein expression and MMP2 secretion. Conclusion Furin siRNA can effectively inhibit the growth of human cervical cancer HeLa cells and the metastasis of tumor cells. The mechanism may be related to the decrease of the expression of MT1-MMP and the synthesis of MMP2.
