目的:优选丹蒲颗粒中丹参和赤芍的醇提取方法及工艺条件。方法:采用正交试验设计考察不同影响因素和水平对芍药苷和丹参酮ⅡA提取率的影响。结果:乙醇回流法芍药苷和丹参酮ⅡA的提取率高于渗漉法和超声法,正交试验筛选出最佳工艺为:8倍量的80%乙醇,回流提取1.5 h,共提取3次。经3批试验验证,对芍药苷和丹参酮ⅡA的提取率分别达到91.2%±4.6%,89.6%±5.2%。结论:优选的醇提工艺简单、高效,对芍药苷和丹参酮ⅡA的提取率高。
Objective: To optimize the extraction method and technological conditions of Salvia miltiorrhiza and Radix Paeoniae Alba in Danpu granule. Methods: The effects of different factors and levels on the extraction rate of paeoniflorin and tanshinone ⅡA were investigated by orthogonal design. Results: The extraction rates of paeoniflorin and tanshinone ⅡA in ethanol reflux method were higher than those of percolation method and ultrasonic method. The optimum technological conditions were as follows: 8 times of 80% ethanol and 1.5 h of reflux extraction. After three batches of experiments, the extraction rates of paeoniflorin and tanshinone ⅡA reached 91.2% ± 4.6% and 89.6% ± 5.2% respectively. Conclusion: The optimized alcohol extraction process is simple and efficient, and the extraction rate of paeoniflorin and tanshinone Ⅱ A is high.
