目的探讨铁皮石斛抑制人鼻咽癌CNE2细胞增殖与诱导凋亡的生物学效应及可能的分子机制。方法分别采用MTT、光学显微镜、流式细胞仪与Western blot检测新鲜铁皮石斛提取物抑制CNE2细胞增殖与诱导凋亡作用及Bcl-xL,mcl-1、Caspase-3、Caspase-8和Caspase-9蛋白的表达,了解铁皮石斛对鼻咽癌细胞的作用及机理。结果经MTT法检测显示铁皮石斛提取物对CNE2有明显抑制作用;经流式细胞仪检测显示,与对照组相比石斛提取物对CNE2细胞株有明显促进细胞凋亡作用(P<0.01),而且有明显的时间依赖性及浓度依赖性。Western blot结果显示:Bcl-xL和Mcl-1表达均下降,Caspase-8、Caspase-9有明显的表达;而Caspase-3也有表达。提示石斛能抑制Bcl-xL和Mcl-1的表达,并可能与线粒体通路的细胞凋亡及死亡通路的细胞凋亡有关。结论铁皮石斛具有抑制人鼻咽癌CNE2细胞增殖和诱导凋亡的作用,其机制可能与Bcl-xL,Mcl-1蛋白下调、促进Caspase-3的活化等有关,并可能与死亡受体通路的细胞凋亡和线粒体通路的细胞凋亡均有关。
Objective To investigate the biological effects and possible molecular mechanisms of Dendrobium officinale on inhibiting the proliferation and inducing apoptosis of human nasopharyngeal carcinoma CNE2 cells. Methods MTT, light microscopy, flow cytometry and Western blot were used to detect the proliferation and apoptosis of CNE2 cells and the effects of extract of Dendrobium candidum on Bcl-xL, mcl-1, Caspase-3, Caspase-8 and Caspase-9 Protein expression, understand the role of Dendrobium officinale on nasopharyngeal carcinoma cells and mechanism. Results MTT assay showed that extract of Dendrobium candidum had a significant inhibitory effect on CNE2. The results of flow cytometry showed that the extract of Dendrobium officinale significantly inhibited the apoptosis of CNE2 cells (P <0.01) compared with the control group, And there is a clear time-dependent and concentration-dependent. The results of Western blot showed that the expressions of Bcl-xL and Mcl-1 were decreased, Caspase-8 and Caspase-9 were obviously expressed, while Caspase-3 was also expressed. Dendrobium can inhibit the expression of Bcl-xL and Mcl-1, and may be related to mitochondrial apoptosis and death pathway apoptosis. Conclusion Dendrobium candidum could inhibit the proliferation and induce the apoptosis of human nasopharyngeal carcinoma CNE2 cells. The mechanism may be related to the down-regulation of Bcl-xL and Mcl-1 protein and the activation of Caspase-3, which may be related to the death receptor pathway Apoptosis and mitochondrial pathway apoptosis are related.
