目的探讨体外缺氧条件下姜黄素对人胰腺癌细胞PANC-1中缺氧诱导因子-1α(HIF-1α)的抑制作用和机制。方法缺氧条件下体外培养PANC-1,RT-PCR检测不同剂量的姜黄素作用24h后HIF-1αmRNA表达的变化;ELISA检测处理后PANC-1细胞中HIF-1α蛋白的表达水平。结果 PANC-1细胞在0.1、1、10、100μmol/L姜黄素处理后,HIF-1α蛋白水平与对照组比较明显降低(F=138.43,P<0.01),且有明显的剂量依赖性,随剂量的增大而降低,但各剂量组HIF-1α的mRNA水平与对照组比较无显著性差异。结论缺氧条件下,姜黄素抑制人胰腺癌PANC-1细胞HIF-1α的转录活性,是通过抑制HIF-1α蛋白的合成来调控的。
Objective To investigate the inhibitory effect of curcumin on hypoxia inducible factor-1α (HIF-1α) in human pancreatic cancer cell line PANC-1 in vitro and its mechanism. Methods PANC-1 was cultured in vitro under hypoxia. The expression of HIF-1αmRNA was detected by RT-PCR after 24h of different doses of curcumin. The expression of HIF-1αprotein in PANC-1 cells was detected by ELISA. Results After treated with 0.1, 1, 10, 100 micromol / L curcumin, the protein level of HIF-1α in PANC-1 cells was significantly lower than that in the control group (F = 138.43, P <0.01) The dose of HIF-1α mRNA was not significantly different from that of the control group. Conclusion Under hypoxia conditions, curcumin inhibits the transcriptional activity of HIF-1α in human pancreatic cancer PANC-1 cells by inhibiting the synthesis of HIF-1α protein.
