目的评价DNA含量(DI)、倍体、S期细胞比例(SPF)、银染核仁形成区(AgNOR)、增殖细胞核抗原(PCNA)及细胞凋亡指数(APO)在口腔粘膜癌前病变癌变监测中的价值及临床应用意义。方法应用流式细胞仪(FCM)、免疫组化等方法对DMBA致地鼠颊囊粘膜癌前病变及癌变过程中的DNA含量、倍体、SPF、AgNOR及APO进行定量检测。结果在Ⅰ—Ⅳ组间(Ⅰ组为正常颊囊粘膜、Ⅱ组为上皮单纯增生、Ⅲ组为上皮异常增生、Ⅳ组为癌肿组)DNA含量、异倍体出现率、SPF、AgNOR计数及PCNA表达依次递增;APO指数随Ⅰ—Ⅲ组依次升高,Ⅳ组时下降。多元逐步判别分析,建立判别函数方程,回代判别准确率为90%。结论本实验结果提示综合监测为口腔粘膜癌前病变癌变的细胞动力学的动态规律的观察提供有益的资料,并对进一步从分子生物学水平研究癌前病变及其癌变将具有重要价值。
Objective To evaluate DNA content (DI), ploidy, S phase cell ratio (SPF), AgNOR, proliferating cell nuclear antigen (PCNA) and apoptotic index (APO) in oral precancerous lesions The value of monitoring and clinical application significance. Methods Quantitative detection of DNA content, ploidy, SPF, AgNOR, and APO in precancerous and carcinogenic lesions of murine cheek pouch mucosa were performed by flow cytometry (FCM) and immunohistochemistry. Results DNA content, aneuploid appearance, SPF, AgNOR counts were observed between I-IV groups (group I was normal cheek pouch mucosa, group II was epithelial hyperplasia, group III was epithelial dysplasia, and group IV was cancer). And the expression of PCNA increased in turn; the APO index increased with the I-III group, and decreased with the IV group. Multivariate stepwise discriminant analysis established the discriminant function equation, and the accuracy rate of back-generation discrimination was 90%. Conclusion The results of this experiment suggest that comprehensive monitoring provides useful information for observing the dynamics of cellular dynamics in the premalignant lesions of the oral mucosa, and will be of great value in the further study of precancerous lesions and their canceration from the molecular biology level.
