目的评价mPEG-PLGA-mPEG(PELGE)空白纳米粒的体外免疫性。方法采用定量酶联检测与PELGE纳米粒共同培养后的THP-1源巨噬细胞细胞因子IL-6、IL-1β和TNF-α的释放量。结果 9种PELGE材料所制备的纳米粒中,9号材料(PEG2000、5%、LA/GA=5:5)可以导致THP-1源巨噬细胞IL-6分泌量显著高于阴性对照组和PLGA纳米粒组。结论 9号材料可能有潜在的免疫毒性,而其余8种材料具有较好的免疫相容性。
Objective To evaluate the in vitro immunity of mPEG-PLGA-mPEG (PELGE) blank nanoparticles. Methods Quantitative enzyme-linked immunosorbent assay (ELISA) was used to detect the release of cytokines IL-6, IL-1β and TNF-α in THP-1-derived macrophages co-cultured with PELGE nanoparticles. Results Among 9 kinds of PELGE nanoparticles, the amount of IL-6 secreted by THP-1-derived macrophages was significantly higher than that of the negative control and the No.9 material (PEG2000, 5%, LA / GA = 5: 5) PLGA nanoparticle group. Conclusion No. 9 material may have potential immunotoxicity, while the remaining 8 kinds of materials have good immunocompatibility.
