目的:探讨归芪胶囊对家兔心肌缺血-再灌注损伤(MIRI)保护作用的可能机制。方法:青紫蓝家兔45只,用随机数字表法分为5组,分别为假手术组、模型组、归芪胶囊小剂量组、归芪胶囊大剂量组和地奥心血康对照组。采用冠状动脉前降支结扎法复制MIRI损伤动物模型,实验过程中动态观察心电图(ECG),以家兔血浆内皮素(ET)、血清一氧化氮(NO),心肌钠泵(Na+-K+-ATPase)和钙泵(Ca2+-Mg2+-ATPase)为指标进行实验观察,用SPSS10.0统计软件进行数据分析。结果:归芪胶囊对MIRI损伤家兔心电图ST段抬高后的恢复性下降,血清NO的升高,血浆ET的降低,心肌Na+-K+-ATPase和Ca2+-Mg2+-ATPase的升高有显著作用(P<0.01)。结论:归芪胶囊通过对血浆ET,血清NO,心肌Na+-K+-ATPase和Ca2+-Mg2+-ATPase含量的影响,对再灌注心肌损伤有保护作用。
Objective: To explore the possible mechanism of Guiqi Capsule on myocardial ischemia-reperfusion injury (MIRI) in rabbits. Methods: Forty-five cyanotic blue rabbits were randomly divided into 5 groups: sham operation group, model group, Guiqi capsule low-dose group, Guiqi capsule high-dose group, and Diaoxin blood control group. The animal model of MIRI injury was duplicated by ligation of the anterior descending coronary artery. The electrocardiogram (ECG) was dynamically observed during the experiment. Rabbit plasma endothelin (ET), serum nitric oxide (NO), and myocardial sodium pump (Na+-K+- ATPase) and calcium pump (Ca2+-Mg2+-ATPase) were used as indicators for experimental observation. SPSS10.0 statistical software was used for data analysis. RESULTS: Guiqi capsules had a significant effect on the restoration of ST-segment elevation, the elevation of serum NO, the decrease of plasma ET, and the increase of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in MIRI-injured rabbits. (P<0.01). Conclusion: Guiqi capsule can protect reperfusion injury of myocardium from plasma ET, serum NO, myocardial Na+-K+-ATPase and Ca2+-Mg2+-ATPase.
