目的观察脾虚证大鼠唾液淀粉酶分泌情况及四君子汤治疗作用。方法雄性SD大鼠随机分为正常组、模型组,模型组,大鼠皮下注射利血平,空白组大鼠注射等量生理盐水,共8d;造模结束后将模型组分为中药组和模型组,中药组给予灌服四君子汤,模型组和正常组给予灌服等量的双蒸水,灌服4周后进行取材。麻醉动物,取下左侧腮腺,缝合伤口,等大鼠稍清醒时用10%冰乙酸溶液20μL刺激大鼠舌尖,1次/min,共刺激30min;处死,取下右侧腮腺。检测腮腺淀粉酶活性、电镜下观察腮腺内酶原颗粒数量、腮腺组织病理。结果模型组大鼠酸刺激后腮腺内残余的淀粉酶活性和酶原颗粒高于正常组大鼠,经四君子汤治疗后脾虚大鼠的唾液淀粉酶的分泌基本恢复正常,各组腮腺组织没有明显病理改变。结论四君子汤对利血平所致脾虚大鼠唾液淀粉酶分泌障碍有明显治疗作用,机制与腮腺组织病理学无关。
Objective To observe the salivary amylase secretion in spleen deficiency rats and the therapeutic effect of Sijunzi Decoction. Methods Male Sprague-Dawley rats were randomly divided into normal group, model group, model group, rats injected subcutaneously with reserpine, and rats in the blank group were injected with the same amount of saline for 8 days. After the model was established, the model group was divided into the traditional Chinese medicine group The model group and the traditional Chinese medicine group were administrated with Sijunzi Decoction. The model group and the normal group were given the same amount of double distilled water. After 4 weeks, the rats were drawn. Animals were anesthetized, the left parotid gland was removed, and the wounds were sutured. When the rats were slightly awake, the tongue of rat was stimulated with 20 μL of 10% glacial acetic acid for 1 min / min and co-stimulated for 30 min. The activity of amylase in parotid gland was detected. The number of zymogen granules in parotid gland and the pathology of parotid gland were observed under electron microscope. Results After acid stimulation, the residual amylase activity and zymogen granules in the parotid gland of rats in model group were higher than those in normal rats. The salivary amylase secretion in spleen deficiency rats returned to normal after treatment with Sijunzi Decoction, and there was no significant difference in parotid gland tissues Pathological changes. Conclusion Sijunzi Decoction has a significant therapeutic effect on salivary amylase secretion in rats with reserpine-induced spleen deficiency. The mechanism is independent of the pathology of the parotid gland.
