目的:优化小菜蛾抗菌肽(CA)的纯化工艺。方法:以总多肽含量、CA含量及Hep G2细胞抑制率为评价指标,选择溶剂类型、提取次数和溶剂用量为考察因素,采用L9(34)正交试验法优选小菜蛾抗菌肽最佳纯化工艺。结果:最优的纯化工艺条件为A3B3C2,即采用p H 6.5磷酸盐缓冲液纯化3次,每次5倍量。结论:该工艺可用于小菜蛾抗菌肽的纯化,其工艺稳定可行,为其新药产业化提供参考。
Objective: To optimize the purification process of antibacterial peptide (CA) in Plutella. Methods: The total peptide content, CA content and inhibition rate of Hep G2 cells were used as evaluation indexes, the type of solvent, the number of extraction and the amount of solvent were selected as the investigation factors. The optimal purification process of antibacterial peptide of Plutella xylostella was optimized by L9 (34) orthogonal test . RESULTS: The optimal purification conditions were A3B3C2, which was purified three times with p H 6.5 phosphate buffer five times each. Conclusion: The process can be used for the purification of antibacterial peptides in Plutella xylostella. The technology is stable and feasible, which provides a reference for the industrialization of new drug.
