本文观察了小鼠肺腺癌LA795母系及不同转移潜能克隆株LA1、LA5细胞其胞内[Ca ̄(2+)]及诱导分化剂维胺酸和二甲基亚砜对其影响。发现母系与克隆株细胞间,其胞内静息[Ca ̄(2+)]确有差异(P<0.01)。中转移LA5克隆株胞内[Ca ̄(2+)]最高,不转移LA1克隆株最低,母系LA705胞内[Ca ̄(2+)]居中。在克隆株细胞中,胞内[Ca ̄(2+)]与其转移潜能成正比。维胺酸作用不同时间,LA795、LA1、LA5胞内[Ca ̄(2+)]先下降,然后逐渐恢复用药前水平,最终至胞内[Ca ̄(2+)]升高。二甲基亚砜作用后,引起胞内[Ca ̄(2+)]一过性升高,然后下降最后又升高。二者均终至所用细胞内[Ca ̄(2+)]升高,提示其诱导分化作用过程中,[Ca ̄(2+)]变化起作用,胞内[Ca ̄(2+)]与肿瘤转移性有一定关系。
In this study, we investigated the effects of levofloxacin and dimethyl sulfoxide (DMSO) on the intracellular [Ca ~ (2+)] and the differentiation of mouse LA795 breast cancer cells and different metastatic potential clones LA1 and LA5. It was found that there was a significant difference in intracellular resting [Ca ~ (2+)] between maternal and cloned cells (P <0.01). LA5 clones had the highest intracellular [Ca ~ (2+)], the lowest LA1 clones without metastasis, and the intracellular [Ca ~ (2+)] in maternal LA705. Intracellular [Ca ~ (2+)] is directly proportional to its metastatic potential in clonal cells. At different time points, the intracellular [Ca ~ (2+)] of LA795, LA1 and LA5 decreased first and then gradually recovered to the level of pretreatment, finally the intracellular [Ca ~ (2+)] increased. Dimethyl sulfoxide, caused intracellular [Ca ~ (2+)] transiently increased, and then decreased and finally increased. Both of them eventually reach the level of [Ca 2+] in the cells used, which suggests that [Ca 2+] plays a role in the process of differentiation induction.
