目的观察紫杉醇对人乳腺癌细胞株MCF-7的凋亡诱导作用,探讨Survivin在此过程中的作用。方法将不同浓度的紫杉醇作用于MCF-7细胞,噻唑蓝(MTT)比色法检测紫杉醇作用的时间效应和剂量效应;用光镜、Hoechst33258荧光染色、流式细胞仪观察细胞的凋亡变化;用逆转录-聚合酶链反应(RT-PCR)、Western blot法观察在紫杉醇作用过程中Survivin的mRNA和蛋白水平的变化。结果细胞生长抑制率呈现一定的时间、剂量依赖性。较高浓度的紫杉醇处理MCF-7细胞48 h后可观察到凋亡。而在各种浓度紫杉醇作用过程的早期(6 h内),Survivin转录和蛋白表达水平明显一过性升高。结论紫杉醇可诱导乳腺癌MCF-7细胞凋亡;而紫杉醇作用过程的早期Survivin表达的增加,极可能有利于肿瘤细胞逃避紫杉醇诱导的细胞凋亡,增加肿瘤细胞耐药的机会。
Objective To observe the apoptosis-inducing effect of paclitaxel on human breast cancer cell line MCF-7 and to explore the role of Survivin in this process. Methods Different concentrations of paclitaxel were applied to MCF-7 cells. The time effect and dose effect of paclitaxel were detected by MTT colorimetric assay. The changes of apoptotic cells were observed by fluorescence microscope and Hoechst 33258 fluorescence staining. Survivin mRNA and protein levels were examined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting. Results The cell growth inhibition rate showed a certain time and dose dependent. Apoptosis was observed in MCF-7 cells treated with paclitaxel at a higher concentration for 48 h. Survivin transcription and protein expression increased transiently in the early stage (within 6 hours) of various concentrations of paclitaxel. Conclusion Paclitaxel can induce apoptosis of MCF-7 breast cancer cells. However, the increased expression of Survivin in the early stage of paclitaxel treatment is likely to help tumor cells escape paclitaxel-induced apoptosis and increase the chances of tumor cell resistance.
