HRAD9是从人脑基因组文库中获取的与酵母菌G2细胞周期检控基因RAD9有高度同源性的基因。功能实验显示,HRAD9能够部分弥补酵母菌中RAD9功能的不足。HRAD9可能具有肿瘤抑制基因的作用,该研究利用SPM方法获得了HRAD9的CpG岛,该序列覆盖HRAD9全长cDNA的5’端非翻译区的79个核苷酸(包括第一个ATG)和部分调控序列(包括起动子),并对该序列进行了详细分析。结果显示,HRAD9的起动子不含TATA盒,但含有GC盒和CCAAT盒。特别令人感兴趣的是,这段调控序列内有CREB,WT1和SRF等调控因子的结合位点。我们认为,这些位点可能是HRAD9与cAMP信号传递系统,WT1肿瘤抑制系统和SRF多功能调控系统的联络环节。
HRAD9 is a gene from the human brain genomic library highly homologous to the yeast cell cycle check gene RAD9 of yeast G2. Functional experiments show that HRAD9 partially compensates for the lack of function of RAD9 in yeast. HRAD9 may have the function of tumor suppressor gene. The study obtained the CpG island of HRAD9 by SPM method, which covers 79 nucleotides (including the first ATG) and part of the 5 ’untranslated region of HRAD9 full-length cDNA Regulatory sequences (including promoters), and the sequence was analyzed in detail. The results showed that the promoter of HRAD9 does not contain the TATA box, but contains the GC box and the CCAAT box. Of particular interest is the presence of regulatory elements such as CREB, WT1 and SRF within this regulatory sequence. In our opinion, these sites may be the link between HRAD9 and cAMP signaling system, WT1 tumor suppressor system and SRF multifunctional control system.
