目的:研究重组血管基膜衍生多功能肽VBMDMP的抗人结肠癌作用。方法:体外培养人结肠癌细胞系(SW480)细胞和中国仓鼠卵巢细胞系(CHO-K1)细胞,采用MTT比色法检测重组VBMDMP对SW480细胞选择性抑制增殖作用;胎盼蓝染色细胞计数法检测重组VBMDMP对SW480细胞和CHO-K1细胞生长曲线的影响;人结肠癌(SW480)细胞裸鼠异种移植瘤模型治疗实验观测重组VBMDMP体内抗人结肠癌作用。结果:重组血管基膜衍生多功能肽具有选择性抑制体外培养人结肠癌细胞系SW480细胞增殖和生长作用,呈剂量依赖性。而对CHO-K1细胞的增殖活性影响小。重组血管基膜衍生多功能肽对SW480细胞裸鼠异种移植瘤生长具有显著抑制作用,呈剂量依赖性。1 mg/kg、5 mg/kg和10 mg/kg重组VBMDMP的肿瘤生长抑制率分别为:63%,79%和83%;瘤重抑制率分别为:62%,66%和75%。其中10 mg/kg重组VBMDMP的肿瘤生长抑制率和瘤重抑制率均接近100 mg/kg环磷酰胺(CTX),高于20 mg/kg血管生成抑制剂烟曲霉素衍生物TNP-470。结论:重组血管基膜衍生多功能肽对人结肠癌细胞增殖和生长具有显著抑制作用。
Objective: To study the anti-human colon cancer effect of recombinant vascular basement membrane-derived multifunctional peptide VBMDMP. Methods: Human colon cancer cell line (SW480) and Chinese hamster ovary cell line (CHO-K1) were cultured in vitro. MTT assay was used to detect the inhibitory effect of recombinant VBMDMP on SW480 cells. The treatment of human colon cancer (SW480) xenograft model in nude mice was used to observe the anti-human colon cancer effect of recombinant VBMDMP in vivo. Results: The recombinant vascular basement membrane-derived multifunctional peptide could selectively inhibit the proliferation and growth of human colon cancer cell line SW480 in a dose-dependent manner. While little effect on the proliferation activity of CHO-K1 cells. Recombinant vascular basement membrane derived multi-functional peptide on SW480 cells in nude mice xenograft tumor growth was significantly inhibited in a dose-dependent manner. The tumor growth inhibition rates of 1 mg / kg, 5 mg / kg and 10 mg / kg recombinant VBMDMP were 63%, 79% and 83%, respectively. The tumor weight inhibition rates were 62%, 66% and 75% respectively. Among them, 10 mg / kg recombinant VBMDMP had a tumor growth inhibition rate and tumor weight inhibition rate close to 100 mg / kg cyclophosphamide (CTX), higher than the 20 mg / kg angiogenesis inhibitor fumagillin derivative TNP-470. Conclusion: The recombinant vascular basement membrane-derived multi-functional peptide has a significant inhibitory effect on the proliferation and growth of human colon cancer cells.
