目的:考察酸碱对药大黄与附子在大黄附子汤中配伍后酸、碱性成分的变化情况。方法:采用高效液相色谱法对大黄、附子单味药提取液、大黄附子对药提取液及全方提取液中蒽醌类成分和乌头生物碱类成分的提取量进行了测定。结果:对大黄而言,蒽醌类成分的提取量随着单药、对药及全方的配伍变化,水煎滤过、水煎离心逐渐减低,乙醇回流蒽醌类成分的提取总量逐渐升高。水煎煮单药与对药、对药与全方之间的变化趋势均明显。对附子而言,水煎煮只检测到单酯型乌头生物碱,乙醇提取既可检测到单酯型乌头生物碱,又可检测到双酯型乌头生物碱;乌头生物碱的提取量随着单药、对药及全方的配伍变化,水煎滤过、水煎离心逐渐减低,乙醇回流变化不明显;水煎煮单药与对药、对药与全方之间的变化趋势均明显。结论:大黄、附子单煎和乙醇回流,大黄、附子对药煎煮和乙醇回流,大黄附子汤全方煎煮和乙醇回流,酸、碱性成分的提取量发生有规律的变化。
