目的探讨多巴胺(DA)对多巴胺能神经母细胞瘤细胞SH-SY5Y的神经毒性作用及可能机制。方法应用MTT法检测DA对SH-SY5Y毒性作用,通过吉姆萨染色、Hoechst 33258标记、透射电镜、DNA ladder以及TUNEL检测DA诱导多巴胺能神经细胞凋亡形态结构及化学特征。结果MTT法检测到DA直接造成多巴胺能神经母细胞损伤,并呈浓度依赖性,有统计学意义(P<0.05);吉姆萨染色、Hoechst33258标记、FACS、电镜、DNA ladder以及TUNEL均检测到细胞凋亡形态及化学特征。结论DA在体外可以造成DA能神经母细胞SH-SY5Y损伤,可能是通过凋亡机制造成细胞丢失的。
Objective To investigate the neurotoxic effect of dopamine (DA) on SH-SY5Y cells in dopaminergic neuroblastoma cells and its possible mechanism. Methods The toxic effects of DA on SH-SY5Y were detected by MTT assay. The morphological and chemical characteristics of dopaminergic neuronal apoptosis were detected by Giemsa staining, Hoechst 33258 labeling, transmission electron microscopy, DNA ladder and TUNEL. Results MTT assay showed that dopaminergic neuroblasts were directly damaged by DA in a concentration-dependent manner (P <0.05). Cells were detected by Giemsa staining, Hoechst33258 labeling, FACS, electron microscopy, DNA ladder and TUNEL Apoptosis morphology and chemical characteristics. Conclusion DA can cause SH-SY5Y injury in DA neuroblasts in vitro, which may be caused by the apoptosis mechanism.
