目的 观察含日本血吸虫候选疫苗分子谷胱甘肽S -转移酶 (Sj2 6 )和天冬酰胺肽链酶 (Sj32 )的真核载体 pBK-CMV -Sj2 6 /Sj32诱导小鼠的免疫应答情况。 方法 将质粒按 5 0 μg/只在 0w ,3w ,5w免疫小鼠股四头肌 ,第一次免疫后取小鼠脾细胞作T淋巴细胞转化实验 ;在 0w、3w和 6w经小鼠尾静脉取血 ,ELISA检测血清中抗体效价及阳性反应率。结果 淋巴细胞转化实验示血吸虫成虫抗原能特异性刺激免疫鼠脾细胞 ;第一次免疫后特异性抗体效价开始升高。抗体最高效价为 1∶2 0 ,3次免疫后有 91.7%小鼠血清呈阳性反应。结论 真核载体 pBK -CMV -Sj2 6 /Sj32可诱导小鼠体液及细胞免疫应答
Objective To observe the immune response induced by the eukaryotic vector pBK-CMV-Sj26 / Sj32 containing Schistosoma japonicum vaccine candidate glutathione S-transferase (Sj26) and asparagine peptide chain enzyme (Sj32). Methods Plasmids were immunized with quadriceps muscle at 0, 3 and 5 weeks at 0, 3 and 5 weeks respectively. After the first immunization, mouse splenocytes were transformed into T lymphocytes. At 0w, 3w and 6w, Blood samples were taken from the venous blood by ELISA, and the antibody titers and positive reaction rates in serum were detected by ELISA. Results Lymphocyte transformation assay showed that the adult schistosome antigen could specifically stimulate the splenocytes of immunized mice. The titer of specific antibody began to increase after the first immunization. The highest titer of the antibody was 1:20, and 91.7% of the mice were positive after 3 immunizations. Conclusion The eukaryotic vector pBK-CMV-Sj26 / Sj32 induces humoral and cellular immune responses in mice
