通过对不同田间温度下的油菜花蕾大小、冷处理时间、更换培养液和培养密度等影响因素试验,建立并优化了长江下游生态型甘蓝型油菜高效小孢子培养体系。结果表明,适宜的花蕾大小与田间温度有密切关系,随着温度升高,应适当降低选取花蕾的长度;冷处理对特定的基因型材料的出胚有效,总体影响不大;在高浓度蔗糖的NLN液体培养基培养2 d后换NLN13培养液后可使材料的出胚数升高;培养密度以1 ml培养液含0.5个花蕾最为合适。
Through experiments on influencing factors such as rape bud size, cold treatment time, medium replacement and culture density under different field temperatures, an efficient microspore culture system of eco-type Brassica napus in the lower reaches of the Yangtze River was established and optimized. The results showed that the suitable flower bud size was closely related to field temperature. With the increase of temperature, the length of buds should be appropriately reduced. Cold treatment had no effect on the embryogenesis of specific genotypes. In the high concentration of sucrose NLN liquid medium for 2 d after the exchange of NLN13 broth can make the embryo of embryos increased; culture density to 1 ml culture medium containing 0.5 buds most appropriate.
