目的探讨冬凌草甲素(Ori)对白血病Molt-4细胞致凋亡作用及其可能的机制。方法将不同浓度的Ori(2.5、5、10、20、40μmol/L)作用于Molt-4细胞。采用甲基噻唑基四唑(MTT)法检测细胞增殖,流式细胞术检测细胞凋亡,电子显微镜观察细胞凋亡超微结构的变化,Western blot方法分析凋亡相关蛋白及Caspase-3表达的变化。结果 Ori可抑制Molt-4细胞的生长及诱导凋亡,并有时间-剂量依赖性;Ori可时间依赖性的下调抗凋亡蛋白Bcl-2的表达,上调促凋亡蛋白Bax和Bim表达以及活化Caspase-3。结论 Ori可诱导Molt-4细胞凋亡,其机制可能与调节Bcl-2家族蛋白及活化Caspase-3有关。
Objective To investigate the apoptosis of Molt-4 cells induced by Oridonin and its possible mechanism. Methods Different concentrations of Ori (2.5, 5, 10, 20 and 40 μmol / L) were applied to Molt-4 cells. Cell proliferation was detected by methylthiazolyltetrazolium (MTT) assay. Apoptosis was detected by flow cytometry. Apoptosis ultrastructure was observed by electron microscopy. Apoptosis-related protein and Caspase-3 expression were analyzed by Western blot Variety. Ori inhibited the growth and induced apoptosis of Molt-4 cells in a time-and dose-dependent manner. Ori down-regulated the expression of anti-apoptotic protein Bcl-2 and the expression of pro-apoptotic proteins Bax and Bim in time-dependent manner Activation of Caspase-3. Conclusion Ori can induce apoptosis in Molt-4 cells. The mechanism may be related to the regulation of Bcl-2 family proteins and the activation of Caspase-3.
