目的:探讨血管内皮细胞生长因子C在颞下颌关节盘前移位后髁突软骨中的表达及意义。方法:采用日本大耳白兔60只,48只用弹力丝牵拉关节盘前移,建立关节盘前移模型,分别于术后1、2、4、8周处死12只,12只空白对照组同期处死,取出颞下颌关节标本。利用基因芯片技术进行筛选,实时荧光定量PCR进行验证和SP免疫组化技术检测不同组内髁突软骨中VGEF-C的表达与分布。采用SPSS19.0软件包对每组样本实验组和对照组数据进行配对t检验。结果:颞下颌关节盘前移位后,在基因水平上VEGF-C相比于正常对照组均有不同程度变化,早期VEGFC表达减少,第2周时降至最低水平,差异显著(P<0.05);第8周时,VEGF-C表达有所增加。镜下观察可见VEGF-C在髁突软骨增殖层、肥大层及钙化软骨层中均有表达,1周组阳性表达主要见于增殖深层和肥大层,钙化软骨层也有弱阳性表达。2周组和4周组阳性表达主要位于肥大层,且在钙化软骨层中表达逐渐增强。8周组肥大层有阳性表达,钙化软骨层未见明显阳性表达,8周组与对照组相比有显著差异(P<0.05)。结论:关节盘前移位后早期髁突软骨VEGF-C基因的转录与表达有所下降,而在后期升高,可能参与关节盘移位后髁突软骨的改建。
Objective: To investigate the expression and significance of vascular endothelial growth factor C in condylar cartilage after temporomandibular joint disc displacement. Methods: Sixty Japanese white rabbits were used. 48 rabbits were pulled forward with stretch yarn to establish an anterior intervertebral disc model. Twelve rabbits were killed at 1, 2, 4 and 8 weeks after operation, and 12 rabbits were blank control Group died at the same period, remove the temporomandibular joint specimens. The gene chip technology was used for screening, real-time fluorescence quantitative PCR for validation and SP immunohistochemistry to detect the expression and distribution of VGEF-C in condylar cartilage in different groups. The SPSS19.0 software package was used to paired t-test with the data of experimental group and control group. Results: Compared with the normal control group, the expression of VEGF-C in the temporomandibular joint shifted to different degrees at the gene level, the expression of VEGFC decreased in the early stage and dropped to the lowest level in the second week (P <0.05 ); At the 8th week, the expression of VEGF-C increased. Microscopically, VEGF-C was expressed in condylar cartilage proliferative layer, hypertrophic layer and calcified cartilage layer. The expression of VEGF-C in the 1-week group was mainly found in the deep and hypertrophic layers, while the calcified cartilage layer was also weakly positive. The positive expression in the 2-week and 4-week groups mainly located in the hypertrophic layer, and gradually increased in the calcified cartilage layer. 8 weeks group hypertrophy positive expression, calcified cartilage layer no significant positive expression, 8 weeks group compared with the control group were significantly different (P <0.05). CONCLUSION: The transcription and expression of VEGF-C gene in early condylar cartilage decreased after anterior plateau displacement, but increased in late stage, which may be involved in the remodeling of condylar cartilage after disc displacement.
