目的 :探讨白细胞介素 - 6 (IL - 6 )加阿霉素 (ADM )对早幼粒细胞株 (HL - 6 0 )凋亡作用的影响。方法 :采用DNA琼脂糖凝胶电泳 ,光镜形态学观察 ,台盼兰活细胞拒染试验分别检测IL - 6、ADM、及IL - 6加ADM对HL - 6 0细胞凋亡的影响。结果 :①中、高剂量IL- 6抑制细胞凋亡 (P <0 0 5 ) ,低剂量无影响 ,(P >0 0 5 )。②ADM诱导细胞凋亡的作用强度及细胞凋亡百分率依次为 :高剂量 >中剂量 >低剂量。③低剂量IL - 6加ADM组的细胞凋亡百分率与ADM组比较无明显差异 (P >0 0 5 ) ,中、高剂量IL - 6加ADM组的细胞凋亡百分率、梯状电泳带 (Ladder)低于同一时间对照组 (P <0 0 5、P <0 0 1)。结论 :IL - 6加ADM可抑制白血病细胞凋亡 ,提示在应用ADM药时慎用IL - 6。
Objective: To investigate the effect of interleukin-6 (IL-6) plus adriamycin (ADM) on the apoptosis of promyelocyte (HL-60) cells. METHODS: DNA agarose gel electrophoresis, light microscope morphology, and trypan blue exclusion test were used to detect the effects of IL-6, ADM, and IL-6 plus ADM on the apoptosis of HL-60 cells. RESULTS : 1 Medium and high doses of IL-6 inhibited apoptosis (P <0 05), while low dose had no effect (P > 0.05). The intensity of the apoptosis induced by 2ADM and the percentage of apoptosis were as follows: high dose >medium dose >low dose. 3 The percentage of apoptotic cells in the low-dose IL-6 plus ADM group was not significantly different from that in the ADM group (P > 0.05), and the percentage of apoptosis in the medium and high-dose IL-6 plus ADM group and the laddering electrophoresis band ( Ladder) was lower than the control group at the same time (P <0 05, P <0 01). Conclusion: IL-6 plus ADM can inhibit apoptosis of leukemia cells, suggesting that IL-6 should be used with caution in the administration of ADM.
