对Romer实验室提出的多功能柱净化,高效液相色谱方法测定果汁中的展青霉素进行了确认,样品经乙腈提取,多功能柱净化一步完成,以四氢呋喃∶水(0.8∶99.2)为流动相,经C18柱分离后用二极管阵列检测器276nm检测。苹果汁中展青霉素回收率在86%~92%,山楂汁中展青霉素回收率在88%~90%,方法的相对标准偏差(RSD)为5%,检出限为8μg/L。然而,该方法的流动相粘度大,反压高,不利于杂质洗脱。经改进流动相,采用乙腈∶水(4∶96),5min后梯度洗脱可消除样品连续测定的干扰,更适合于实际样品测定。
The determination of patulin in fruit juice by multifunctional column purification and high performance liquid chromatography (HPLC) was carried out in Romer laboratory. The sample was extracted with acetonitrile and purified by multifunctional column. The mobile phase consisted of tetrahydrofuran: water (0.8: 99.2) , After the C18 column with a diode array detector 276nm detection. The recoveries of patulin in apple juice ranged from 86% to 92%. The recoveries of patulin from hawthorn juice ranged from 88% to 90%. The relative standard deviation (RSD) was 5% and the detection limit was 8μg / L. However, this method has a high mobile phase viscosity and a high backpressure, which is detrimental to impurity elution. After the mobile phase was improved, the gradient elution of acetonitrile: water (4:96) after 5 min could eliminate the interference of the continuous determination of the sample, which is more suitable for the actual sample determination.
