目的 :以银杏叶粗提物为原料 ,利用高速逆流色谱法 (HSCCC)分离制备得到白果内酯单体。方法 :采用氯仿 甲醇 水 ( 4∶3∶2 )的溶剂系统 ,待两相溶剂充分混合后 ,临用前将其分开 ,在室温下 ,下相作流动相 ,上相作固定相 ,以 2 .0ml/min的流速 ,由首端向尾端洗脱 ,主机正转 ,转速为 80 0r/min ,固定相保留值为 78% ,进样 2 .0ml,定时收集馏分 ,用TLC鉴定。结果 :经一次HSCCC即可分离制得白果内酯单体 ,TLC方法可用于银杏内酯的定性检测。结论 :本方法简便、快速 ,为中草药有效成分的制备提供了一条新途径
OBJECTIVE: To isolate bilobalide monomer by high speed countercurrent chromatography (HSCCC) using crude extract of Ginkgo biloba as raw material. Methods: The solvent system of chloroform-methanol water (4: 3: 2) was used. After the two-phase solvent was mixed well, it was separated before use. The mobile phase was phase at room temperature, .0ml / min flow rate from the beginning to the end of the elution, the host forward rotation speed of 80 0r / min, stationary phase retention of 78%, injection 2.0ml, regular collection of fractions identified by TLC. Results: The bilobalide monomer could be separated and isolated by a single HSCCC. TLC method could be used for the qualitative detection of ginkgolide. Conclusion: The method is simple and rapid, which provides a new way to prepare the active ingredients of Chinese herbal medicine
