目的:采用亲水作用色谱-蒸发光散射检测器联用法同时测定巴戟天中蔗糖、蔗果三糖、耐斯糖、1F-果呋喃糖基耐斯糖、巴戟甲素5种低聚糖的含量。方法:采用Waters XBridgeTMAmide(4.6 mm×150 mm,3.5μm)亲水性作用色谱柱;以乙腈(A)-0.2%三乙胺溶液(B)为流动相,梯度洗脱;流速0.8 mL·min-1;柱温40℃;以Waters 2424 ELSD检测,漂移管温度90℃,气压275.79 kPa。结果:5种低聚糖分别在2.128~21.28μg(r=0.999 3),1.864~18.64μg(r=0.999 6),1.92~19.2μg(r=0.999 8),1.912~19.12μg(r=0.999 5),2.368~23.68μg(r=0.999 4)线性关系良好,其回收率在92.81%~102.8%(n=6)。结论:该方法简单、准确,具有良好的重复性,为有效评价巴戟天药材的质量提供了分析方法。
OBJECTIVE: Simultaneous determination of sucrose, fructose trisaccharide, nesiritide, 1F-fructo-furanosylstilbestigmine, and 5 kinds of oligo-oligosaccharides of Radix Morinda officinalis by hydrophilic interaction chromatography-evaporative light scattering detector Sugar content. Methods: Waters XBridge TMAmide (4.6 mm × 150 mm, 3.5 μm) hydrophilic column was used. The mobile phase consisted of acetonitrile (A) -0.2% triethylamine (B) -1. The column temperature was 40 ℃. The sample was detected by Waters 2424 ELSD. The drift tube temperature was 90 ℃ and the pressure was 275.79 kPa. Results: The concentrations of five oligosaccharides in the range of 2.128-21.28μg (r = 0.999 3), 1.864-18.64μg (r = 0.999 6), 1.92-19.2μg (r = 0.999 8), 1.912-19.19μg 5) and 2.368 ~ 23.68μg (r = 0.999 4). The recoveries ranged from 92.81% to 102.8% (n = 6). Conclusion: The method is simple, accurate and reproducible. It provides an analytical method for the effective evaluation of medicinal quality of Morinda officinalis.
