目的:研究高糖诱导下,杭白菊黄酮对人脐静脉血管内皮细胞(HUVEC)表达Bax、Bcl-2的影响。方法:将HUVEC分为空白对照组、正常血糖组、高糖刺激组和杭白菊高、中、低剂量组,作用24和48h后,观察各组细胞形态变化,采用MTT法检测细胞生长状况,RT-PCR法检测内皮细胞Bax、Bcl-2的表达。结果:高糖诱导下,杭白菊黄酮可下调Bax的表达,上调Bcl-2的表达,而杭白菊黄酮呈剂量依赖性抑制Bax表达,增强Bcl-2表达。结论:杭白菊黄酮可通过下调高糖诱导的血管内皮细胞Bax表达,上调Bcl-2表达,抑制血管内皮细胞凋亡,在一定程度上有助于防治糖尿病血管病变,为临床应用杭白菊防治糖尿病患者血管并发症提供理论参考。
AIM: To investigate the effects of Chrysosporium flavone on the expression of Bax and Bcl-2 in human umbilical vein endothelial cells (HUVECs) induced by high glucose. Methods: The HUVECs were divided into blank control group, normal blood glucose group, high glucose stimulation group and Chrysanthemum high, medium and low dose groups for 24 and 48 hours, the morphological changes of the cells in each group were observed, the cell growth was detected by MTT assay, The expression of Bax and Bcl-2 in endothelial cells was detected by RT-PCR. Results: Under the condition of high glucose, Bax could down-regulate the expression of Bax and up-regulate the expression of Bcl-2, while the Chrysanthemum flavonoids could inhibit the expression of Bax in a dose-dependent manner and enhance the expression of Bcl-2. Conclusion: Chrysanthemum flavone can prevent and treat diabetic vascular disease by down-regulating Bax expression induced by high glucose, up-regulating Bcl-2 expression and inhibiting apoptosis of vascular endothelial cells, which is helpful for the clinical application of Chrysanthemum against diabetes Provide a theoretical reference for patients with vascular complications.
