目的探讨细胞周期素D1(cyclinD1)、细胞周期素E(cyclinE)及p27在肾盂移行细胞癌中的表达及意义。方法选择肾盂移行细胞癌手术切除标本63例,WHO(1998)病理分级:G112例、G240例、G311例;UICC(1997)TNM病理分期:Ta4例、T19例、T224例、T317例、T49例。11例因外伤切除正常肾盂组织作为正常对照组。应用免疫组化二步法检测组织中cyclinD1、cyclinE和p27的表达。结果正常肾盂组织中,cyclinD1呈弱表达,cyc-linE不表达,p27呈高表达;肾盂移行细胞癌组织中,cyclinD1与cyclinE表达显著增高(P<0.05),p27表达显著降低(P<0.05)。随病理分级及分期的增高,cyclinD1与cyclinE的阳性表达率升高(P<0.05),p27的阳性表达率下降(P<0.05),且cyclinD1、cyclinE均与p27的表达呈负相关(P<0.05)。结论肾盂移行细胞癌组织中可见cyclinD1、cyclinE的过度表达及p27的失活,三者的联合检测有可能作为判断肾盂移行细胞癌恶性程度和预后的生物学指标。
Objective To investigate the expression and significance of cyclin D1, cyclin E and p27 in renal pelvis and transitional cell carcinoma. Methods Sixty-three cases of resected renal pelvis and transitional cell carcinoma were selected according to WHO (1998) pathological grading: G112 cases, G240 cases and G311 cases; UICC (1997) TNM pathological stage: Ta4 cases, T19 cases, T224 cases, T317 cases and T49 cases . 11 cases of normal renal pelvis due to traumatic resection as a normal control group. The expression of cyclinD1, cyclinE and p27 was detected by immunohistochemical two-step method. Results The expression of cyclinD1 and cyclinE1 in renal pelvis and transitional cell carcinoma was significantly increased (P <0.05) and p27 was significantly decreased (P <0.05) . The positive rate of cyclinD1 and cyclinE increased (P <0.05), the positive rate of p27 decreased (P <0.05), and cyclinD1 and cyclinE were negatively correlated with the expression of p27 (P < 0.05). Conclusions The overexpression of cyclinD1, cyclinE and inactivation of p27 are seen in renal pelvis and transitional cell carcinoma of the renal pelvis. The combined detection of the three may be used as biological indicators to judge the malignancy and prognosis of renal pelvis transitional cell carcinoma.
