摘要[目的]对大豆异黄酮提取纯化的最佳工艺条件及其抗氧化活性进行研究。[方法]通过单因素试验和L9(34)正交试验, 确定提取大豆异黄酮的最佳工艺条件,应用D101大孔树脂技术对提取液进行进一步分离纯化,得出最佳纯化条件,并对纯化得到的染料木苷和大豆苷进行抗氧化活性研究。[结果] 试验得出,提取大豆异黄酮的最佳工艺条件为乙醇浓度70%,料液比1∶15 g/ml,提取时间为3 h,提取温度为60 ℃,最高得率达9.18%;纯化最佳条件为:上柱静态吸附时间5 h,洗脱时间30 min,80%乙醇作为洗脱剂,洗脱流速为l ml/min,并分离纯化得到染料木苷和大豆苷;抗氧化活性研究表明,染料木苷、大豆苷和大豆总黄酮对超氧阴离子自由基和羟自由基均具有清除作用。[结论] 研究对大豆保健食品开发和天然药物研制具有重要意义。

Extraction, Purification Technology and Antioxidation Activities of Soybean Isoflavones

Abstract [Objective] To study the optimum technique conditions and antioxidation activities of extraction and purification of soybean isoflavones. [Method] On the basis of single element experiments and L9(34) orthogonal experiments, the optimum technique conditions for extraction of soybean isoflavone were determined. D101 macroporous resin technology was applied to further purify the liquids. [Result] The optimum extraction conditions were obtained: ethanol concentration 70%, soybean to solvent ratio 1∶15 g/ml, extraction time 3 h, temperature 60 ℃. The extraction yield could be up to 9.18%. Purification process of soy isoflavones in soybean by using D101 macroporous resin, the optimum purification conditions were obtained: absorption time 5 h, elution time 30 min, ethanol concentration 80%, elution speed l ml/min, and obtain high purity of daidzin, genistin monomer. The antioxidation activity was analyzed, and the conclusion revealed that soybean isoflavones possessed superoxide radical and hydroxyl radical scavenging activities.[Conclusion] The study has significance on development of soybean health food and natural medicine.

大豆异黄酮 (soybean isoflavone)是大豆生长过程中形成的主要次生代谢产物之一,具有防治癌症、预防心血管疾病、改善骨质疏松、降低血脂、防止动脉粥样硬化和改善妇女更年期综合症等多种生理功能[1-3]。近年来,大豆异黄酮已被开发成多种保健功能产品。20世纪90年代以来,对大豆异黄酮的提取工艺研究有不少报道[ 4-10],由于大豆中異黄酮的含量较低,提取成本较高且提取工艺复杂,因此高效提取纯化仍是目前提取工艺的一个关键性的问题。为高效利用大豆种子中的大豆异黄酮,笔者对大豆异黄酮的提取、分离和纯化工艺进行了优化,并且分别检测了染料木苷、大豆苷和大豆总黄酮的抗氧化作用,对大豆保健食品开发和天然药物研制具有重要意义。
1材料与方法
1.1材料供试大豆种子晋大70,由山西农业大学农学院大豆育种课题组提供;所用试剂均为分析纯。主要仪器: 紫外分光光度计,真空旋转蒸发仪,高速台式离心机,Brucker600型核磁共振仪。
