目的:研究蛋白激酶CK2α特异性siRNA对人喉癌Hep-2细胞裸鼠移植瘤生长和蛋白激酶CK2α表达的影响。方法:裸鼠皮下种植人喉癌Hep-2细胞,肿瘤长到一定大小时,在肿瘤局部注射蛋白激酶CK2α特异性siRNA表达质粒,观察肿瘤生长情况,应用逆转录-聚合酶链反应(RT-PCR)检测蛋白激酶CK2αmRNA在肿瘤中的表达,应用免疫组织化学方法检测蛋白激酶CK2α蛋白在肿瘤中的表达。结果:转染蛋白激酶CK2α特异性siRNA表达质粒后,裸鼠移植瘤生长缓慢(P<0.01),裸鼠移植瘤中蛋白激酶CK2αmRNA和蛋白表达均较非特异干扰组和空白组明显下降(P<0.05)。结论:蛋白激酶CK2α特异性siRNA表达载体可以下调裸鼠移植瘤中蛋白激酶CK2α表达,抑制肿瘤生长。RNA干扰技术可能成为治疗喉癌的一种新途径。
OBJECTIVE: To study the effect of protein kinase CK2α-specific siRNA on the growth of human transplanted tumor and the expression of protein kinase CK2α in human laryngeal carcinoma Hep-2 cells. Methods: Human laryngeal carcinoma Hep-2 cells were implanted subcutaneously in nude mice. When the tumor grew to a certain size, protein kinase CK2α specific siRNA expression plasmids were injected locally to observe the tumor growth. RT- PCR) was used to detect the expression of protein kinase CK2αmRNA in the tumor. Immunohistochemistry was used to detect the protein kinase CK2αprotein expression in the tumor. Results: After transfection with protein kinase CK2α specific siRNA plasmid, the transplanted tumor in nude mice grew slowly (P <0.01), and the expression of protein kinase CK2α mRNA and protein in nude mice transplanted tumor was significantly decreased compared with non-specific interference group and blank control group (P <0.05). Conclusion: The protein kinase CK2α-specific siRNA expression vector can down-regulate the expression of protein kinase CK2α in tumor xenografts of nude mice and inhibit tumor growth. RNA interference technology may become a new way to treat laryngeal cancer.
