该研究旨在通过探讨蜘蛛香环烯醚萜类成分缬草醛对肠易激综合征(IBS)大鼠结肠中促肾上腺皮质激素释放因子(CRF)、色氨酸羟化酶1(TPH1)mRNA表达及5-羟色胺(5-HT)含量的影响,阐释缬草醛通过5-HT通路治疗IBS的作用机制。将54只SD雄性大鼠随机分为空白对照组、模型组、缬草醛高、中、低剂量组、匹维溴铵组,每组9只。采用孤养结合慢-急性不可预见性应激法建立IBS大鼠模型,免疫组化法测定大鼠结肠CRF表达,RT-PCR法测定结肠中TPH1 mRNA表达,高效液相色谱法测定大鼠结肠中5-HT含量。实验结果显示,孤养结合慢-急性不可预见性应激可造成大鼠结肠中CRF,TPH1 mRNA表达以及5-HT含量升高(P<0.05),缬草醛各剂量组中CRF,TPH1 mRNA及5-HT含量较模型组明显下降(P<0.05)。实验结果表明,肠易激综合征能够导致结肠中CRF,TPH1 mRNA的表达以及5-HT的含量升高,而缬草醛通过降低结肠中CRF,TPH1 mRNA的表达以及5-HT的含量,而改善胃肠道的相关症状。
The purpose of this study was to investigate the effects of valerian on the colon of corticotropin-releasing factor (CRF), tryptophan hydroxylase 1 (TPH1) mRNA expression and 5-hydroxytryptamine (5-HT) content, explaining the mechanism of valeraldehyde treatment of IBS by 5-HT pathway. Fifty-four SD male rats were randomly divided into blank control group, model group, valerian aldehyde high, medium and low dose groups, and piroxicam group, with 9 rats in each group. IBS rat model was established by solitary and slow-acute unpredictable stress, the expression of CRF in colon was determined by immunohistochemical method, the expression of TPH1 mRNA in colon was detected by RT-PCR and the expression of TPH1 mRNA was detected by high performance liquid chromatography In the 5-HT content. The experimental results showed that solitary combined with slow - acute unpredictable stress can cause CRF, TPH1 mRNA expression in rat colon and 5-HT content increased (P <0.05), CRF, TPH1 mRNA And 5-HT levels were significantly lower than the model group (P <0.05). Experimental results show that irritable bowel syndrome can lead to CRF, TPH1 mRNA expression in the colon and 5-HT content increased, while valeraldehyde by reducing the expression of CRF, TPH1 mRNA in the colon and 5-HT content, and Improve the gastrointestinal tract related symptoms.
