利用大鼠胚胎中脑神经细胞进行原代培养(微团试验)研究氟化钠(NaF)的细胞毒性及其对神经元分化的影响。细胞毒性试验提示NaF的半数存活抑制浓度(ICV50)为15.0μg/ml。染毒组的细胞集落形成率降低,细胞体积小,细胞间神经纤维减少等形态学变化,其中前者呈剂量—效应关系,表明NaF可抑制胚胎神经细胞的分化,半数分化抑制深度(ICD50)为3.0μg/ml,半数存活抑制深度与半数分化抑制浓度的比值(V/D)大于5.0。表明抑制细胞分化可能是NaF致发育危害的重要作用机理。
Primary culture of rat embryo midbrain neurons (micellar assay) was used to study the cytotoxicity of sodium fluoride (NaF) and its effect on neuronal differentiation. Cytotoxicity assays suggest that the half-life inhibitory concentration (ICV50) of NaF is 15.0 μg / ml. In the exposure group, morphological changes such as decreased colony formation rate, small cell size and decreased intercellular nerve fibers were observed. The former showed a dose-response relationship, indicating that NaF inhibited the differentiation of embryonic neurons. The ICD50 was 3.0μg / ml, the ratio of the half survival inhibition depth to half differentiation inhibition concentration (V / D) was greater than 5.0. Indicating that inhibition of cell differentiation may be an important mechanism of action of NaF-induced development.
