目的:建立HPLC-电喷雾检测器检测长春瑞滨脂质体中单硬脂酰磷脂酰胆碱(MSPC)含量的方法并验证。方法:采用Waters e2695系列高效液相色谱系统联合电喷雾检测器进行测定。采用Zorbax Eclipse Plus C8色谱柱,流动相为甲醇-水,梯度洗脱,流速1.0 mL·min-1。检测器参数:气压35 psi(house nitrogen),量程200 pA。结果:此方法线性范围为4.8~28.8μg·mL-1(r=0.999 9),重复性、中间精密度RSD均<2.0%,平均回收率为100.43%。结论:本方法可以快速、简便的测定长春瑞滨热敏脂质体中MSPC的含量,灵敏度高,相应一致性好,充分说明了电喷雾检测器适于检测脂质体中的低浓度溶血磷脂。
OBJECTIVE: To establish a method for the determination of monostearyl phosphatidylcholine (MSPC) in vinorelbine liposomes by HPLC-electrospray detector. Methods: Waters e2695 series high performance liquid chromatography coupled with electrospray detector was used for the determination. A Zorbax Eclipse Plus C8 column was used with a mobile phase of methanol-water and gradient elution at a flow rate of 1.0 mL · min-1. Detector parameters: 35 psi (house nitrogen), 200 pA range. Results: The linear range of this method was 4.8 ~ 28.8μg · mL-1 (r = 0.999 9). The repeatability and intermediate precision RSD were all less than 2.0% and the average recovery was 100.43%. Conclusion: This method can be used to determine the content of MSPC in vinorelbine thermosensitive liposomes quickly and easily, with high sensitivity and good consistency. It fully demonstrates that electrospray detector is suitable for detecting low concentration of lysophospholipids in liposomes .
