目的:探讨四逆汤改善缺血心肌能量代谢的作用及其机制。方法:垂体后叶素性小鼠心肌缺血模型,四逆汤(浓度103g生药/L)灌胃,剂量01mL/20gwt·d-1,心肌超微结构用电镜观察分析,心肌ATP含量用高效液相色谱法测定,乳酸浓度用常规生化方法测定。结果:与缺血组相比,四逆汤组缺血心肌线粒体损伤显著减轻(P<005),糖原消耗显著减少(P<005),乳酸浓度显著下降(P<005),心肌营养血流量显著上升(P<001),氧自由基浓度显著降低(P<001)。结论:四逆汤具有显著改善缺血心肌能量代谢的作用,该作用与其增加心肌供血和清除氧自由基有关
Objective: To investigate the effect and mechanism of Sini decoction on energy metabolism of ischemic myocardium. METHODS: Pituitrin-induced mouse myocardial ischemia model, Sini Decoction (concentration of 103 g crude drug/L) was intragastrically administered, and the dose was 01 mL/20gwt·d-1. Myocardial ultrastructure was observed by electron microscopy, and the myocardial ATP content was used. As determined by high performance liquid chromatography, the lactic acid concentration was determined using routine biochemical methods. Results: Compared with the ischemic group, the mitochondrial damage of ischemic myocardium in SND group was significantly reduced (P<0. 05), glycogen consumption was significantly reduced (P<0. 05), and the lactic acid concentration was significantly decreased (P<0. 05), myocardial blood flow significantly increased (P <0 01), oxygen radical concentration was significantly reduced (P <0 01). Conclusion: Sini Decoction can significantly improve the energy metabolism of ischemic myocardium. This effect is related to the increase of myocardial blood supply and the clearance of oxygen free radicals.
