目的:研究复方六月青(CLYQ)对肝星状细胞(HSC)凋亡的影响。方法:采用四甲基噻唑蓝(MTT)法检测CLYQ对细胞的毒性。采用吖啶橙荧光染色和流式细胞术检测CLYQ对HSC凋亡的影响。结果:CLYQ对HSC毒性很低。吖啶橙荧光染色法结果显示,与空白对照组相比较,CLYQ对HSC-T6凋亡细胞比例的影响有统计学差异(P<0.01),其凋亡率细胞比例有明显的剂量依赖性。流式细胞术结果显示,空白对照组凋亡细胞比例为4.82,CLYQ 3.6mg/mL浓度组凋亡细胞比例为10.10,1.8mg/mL浓度组凋亡细胞比例为6.76,0.9mg/mL浓度组凋亡细胞比例为5.18。结论:CLYQ通过诱导HSC细胞凋亡,从而发挥抗肝纤维化作用。
Objective: To study the effect of CLYQ on the apoptosis of hepatic stellate cells (HSC). Methods: Tetramethylthiazolyl blue (MTT) method was used to detect the cytotoxicity of CLYQ. Acridine orange fluorescence staining and flow cytometry were used to detect the effects of CLYQ on HSC apoptosis. Results: CLYQ has low toxicity to HSC. Acridine orange fluorescence staining showed that, compared with the blank control group, the effect of CLYQ on the proportion of apoptotic cells in HSC-T6 was statistically significant (P <0.01), and the percentage of apoptotic cells was significantly dose-dependent. The results of flow cytometry showed that the percentage of apoptotic cells in the blank control group was 4.82, the percentage of apoptotic cells in the group of CLYQ 3.6 mg / mL was 10.10, and the percentage of apoptotic cells in the group of 6.7 mg / mL and 1.8 mg / mL was 6.76 and 0.9 mg / mL The percentage of apoptotic cells was 5.18. Conclusion: CLYQ can induce hepatic fibrosis by inducing HSC cell apoptosis.
