建立了高效阴离子交换色谱法同时测定贼小豆中9种糖类化合物含量的分析方法。用CarboPacTMPA10阴离子交换分离柱和脉冲安培检测器,以0.25 mol/L氢氧化钠和水等度洗脱,流速0.80 mL/min,柱温30℃。结果表明:25 min内能完成9种糖的分离,在0.05~25.0 mg/L范围内呈良好的线性关系,相关系数大于0.9993,检出限为0.05~1.20μg/L,加标回收率为91.66%~103.2%,相对标准偏差RSD小于2.3%(n=6)。方法灵敏度高,前处理简单快速、选择性好、无需衍生,用于郑州地区野生贼小豆中糖类化合物的快速检测,结果较满意。
A high performance anion exchange chromatography was established for simultaneous determination of nine carbohydrates in thief adzuki bean. A CarboPac TMPA10 anion exchange column and a pulsed amperometric detector were used. The eluate was eluted with 0.25 mol / L sodium hydroxide and water at a flow rate of 0.80 mL / min and the column temperature was 30 ℃. The results showed that the separation of nine kinds of sugars could be completed within 25 min, with good linearity in the range of 0.05-25.0 mg / L with the correlation coefficient greater than 0.9993 and the detection limit of 0.05 ~ 1.20 μg / L with the recoveries of 91.66% ~ 103.2%. The relative standard deviation (RSD) was less than 2.3% (n = 6). The method has the advantages of high sensitivity, simple and rapid pretreatment, good selectivity and no need of derivatization. It is used for the rapid detection of carbohydrates in the wild thief bean in Zhengzhou area. The results are satisfactory.
