目的研究Cdc42基因在新疆维吾尔族食管癌组织中的表达并探讨其在维吾尔族食管癌发生、发展中的作用。方法采用逆转录多聚合酶链反应技术检测Cdc42基因在20例食管癌组织及其相应癌旁组织中的表达情况,并与临床病理资料结合分析其意义。结果癌组织与相应癌旁组织相比,在20例癌组织中,有15例表达阳性,占75%;癌组织表达量明显高于癌旁组织的有7例,占35%;其中有淋巴结转移的癌组织中Cdc42 mRNA的表达率为80%(8/10),无淋巴结转移的为60%(6/10);肿瘤浸润至外膜层的表达率为80%(8/10)。结论提示Cdc42基因在转录水平有的异常表达,Cdc42基因在不同分期的癌组织中,Cdc42基因高表达与淋巴结转移和肿瘤浸润深度有关。Cdc42基因在癌组织和不同分期癌组织中表达的改变,提示它所参与的细胞凋亡及细胞周期过程的变化在维吾尔族食管癌的发生、发展、转移、浸润过程中起了一定的作用。
Objective To study the expression of Cdc42 in esophageal carcinoma of Uygur in Xinjiang Uygur and its role in the occurrence and development of Uygur esophageal cancer. Methods The expression of Cdc42 gene in 20 cases of esophageal cancer tissues and corresponding paracancerous tissues was detected by reverse transcription polymerase chain reaction (RT-PCR), and its significance was analyzed with clinicopathological data. Results Of the 20 cases of cancerous tissue, 15 cases were positive in cancerous tissue, accounting for 75%; the expression of cancerous tissue was significantly higher than that in 7 cases of para-cancerous tissue (35%). Among them, there were lymph nodes The rate of Cdc42 mRNA expression was 80% (8/10) and no lymph node metastasis was 60% (6/10). The rate of tumor invasion to the outer membrane was 80% (8/10). The results suggest that Cdc42 gene is abnormally expressed at the transcriptional level. The high expression of Cdc42 gene in different stages of cancerous tissue correlates with the lymph node metastasis and the depth of tumor invasion. The change of Cdc42 gene expression in cancer tissues and different stages of cancer suggests that the changes of apoptosis and cell cycle involved in it play a role in the occurrence, development, metastasis and invasion of Uighur esophageal cancer.
