目的研究真皮成纤维细胞在上皮覆盖过程中细胞因子基因水平的变化。 方法运用人cDNA微阵列技术 ,比较覆盖或不覆盖表皮层的真皮模拟物中成纤维细胞所含的细胞因子mRNA水平的变化。 结果成纤维细胞在 2 6 8个细胞因子基因中表达 6 8个基因 ,在 2 6 5个与细胞间相互作用相关的基因中有 6 4个被表达。第 7d时 ,4个基因上调 ,6个基因下调 ;第 15d时 ,4个基因上调 ,9个基因下调。 结论无表皮覆盖的真皮组织成纤维细胞中趋化因子的mRNA水平增高 ,金属蛋白酶mRNA水平下降 ,这有利于刺激上皮细胞的增殖和移动 ,促进细胞外基质的重塑
Objective To study the changes of cytokine gene level in dermal fibroblasts during epithelial coverage. Methods Human cDNA microarray was used to compare the mRNA levels of cytokines contained in fibroblasts in dermis mulches with and without epidermis. Results Fibroblasts expressed 68 genes in 268 cytokine genes and 6 4 genes in 2650 genes related to cell-cell interaction. On the 7th day, 4 genes were up-regulated and 6 genes down-regulated. On the 15th day, 4 genes were up-regulated and 9 genes were down-regulated. Conclusion The mRNA expression of chemokines in dermal fibroblasts without epidermis is increased and the level of metalloproteinase mRNA is decreased, which is good for stimulating the proliferation and migration of epithelial cells and promoting the remodeling of extracellular matrix
