目的 :观察中草药提取物 MACC5 (为广东医学院肾脏病研究所提取物代号 )对 SD大鼠抗 Thy1.1系膜增生性肾炎实验模型的疗效。方法 :制备兔抗大鼠胸腺细胞免疫血清 (ATS) ,再将 ATS经尾静脉注射给 SD大鼠 ,诱导其发生系膜增生性肾炎 (Ms PGN)后 ,随机分为 3组 ,每组 5只 ;试验组分别经腹腔注射 MACC5 1.0 m L 和 3.0 m L,对照组注射生理盐水 (NS) 1.0 m L ;每日观察其尿量、尿蛋白定量 ,并于第 11天处死各大鼠 ,取肾组织切片观察系膜增殖程度 ,并用免疫组化法检测肾小球系膜区细胞增殖核抗原 (PCNA)的表达。结果 :两试验组 2 4h尿量均多于对照组 (P<0 .0 5 ) ;两试验组尿蛋白定量均少于对照组 (P<0 .0 1) ;两试验组单位肾小球系膜细胞 (GMC)数均少于对照组 (P<0 .0 0 1) ;MACC5 3.0m L试验组肾小球系膜区 PCNA的表达低于对照组 (P<0 .0 5 )。结论 :MACC5对大鼠 Ms PGN实验模型具有利尿、降低尿蛋白和促进肾小球系膜损伤修复的作用。
Objective: To observe the efficacy of the Chinese herbal extract MACC5 (extracted for the Institute of Nephrology, Guangdong Medical College) for the experimental model of anti-Thy1.1 mesangial proliferative nephritis in SD rats. METHODS: Rabbit anti-rat thymocyte immune serum (ATS) was prepared, and ATS was injected into SD rats via tail vein to induce mesangial proliferative nephritis (Ms PGN). The rats were randomly divided into 3 groups with 5 in each group. Only in the experimental group were intraperitoneal injection of MACC5 1.0 m L and 3.0 m L, and the control group was injected with 1.0 m L of normal saline (NS). The urine volume and urinary protein were observed daily, and each rat was sacrificed on the eleventh day. Renal tissue sections were taken to observe the extent of mesangial proliferation, and the expression of cell proliferation nuclear antigen (PCNA) in mesangial area was detected by immunohistochemistry. Results: The urine volume in both groups was higher than that in the control group (P<0.05); the urinary protein quantification was lower in the two experimental groups than in the control group (P<0.01); the two experimental groups were glomeruli The number of mesangial cells (GMC) was less than that of the control group (P<0.01); the expression of PCNA in glomerular mesangial area was lower in MACC5 3.0m L test group than in the control group (P<0.05). Conclusion : MACC5 can diuresis, reduce urinary protein and promote the repair of glomerular mesangial injury in rat Ms PGN experimental model.
