目的:采用高效液相色谱法同时测定康尔心胶囊丹参5种水溶性成分(丹参素钠、原儿茶醛、紫草酸、迷迭香酸、丹酚酸B)。方法:采用Thermo syncronis-C18色谱柱(4.6 mm×250mm,5μm),以甲醇为流动相A,1%冰醋酸为流动相B进行梯度洗脱;检测波长:281nm,流速为1.0ml·min-1,进样量为10μL,结果:丹参素钠、原儿茶醛、紫草酸、迷迭香酸、丹酚酸B 5种成分在55分钟以内基本分离。峰面积(Y)对进样量(X)的标准曲线分别为丹参素钠Y=642.88X+1.98,r=0.9999;原儿茶醛Y=4372213.83X+805.16,r=0.9999;紫草酸Y=1245713.26X+4412.89,r=0.9999;迷迭香酸Y=1713766.30X+1387.01,r=0.9999;丹酚酸B Y=1209485.54X+29336.62,r=0.9998;加样回收率分别为丹参素钠99.8%(RSD 1%);原儿茶醛98.5%(RSD 0.9%)、紫草酸98.3%(RSD 1.6%)、迷迭香酸103.8%(RSD0.7%)、丹酚酸B 98.3%(RSD 0.8%)。结论:该方法操作简便,重复性好,分离效果好,可以作为康尔心胶囊的质量控制。
Objective: To determine the five water soluble constituents of Salvia miltiorrhiza (Danshensu, protocatechuic aldehyde, lithospermic acid, rosmarinic acid and salvianolic acid B) by high performance liquid chromatography (HPLC). METHODS: The mobile phase A was eluted with a gradient of methanol (4.6 mm × 250 mm, 5 μm) using mobile phase A and 1% glacial acetic acid as mobile phase B. The detection wavelength was 281 nm and the flow rate was 1.0 ml · min- 1, the injection volume of 10μL, the results: sodium danshensu, protocatechuic aldehyde, purple oxalic acid, rosmarinic acid, salvianolic acid B 5 kinds of components within 55 minutes basically separated. The standard curves of peak area (Y) and injection volume (X) were respectively sodium danshensu Y = 642.88X + 1.98, r = 0.9999; protocatechuicide Y = 4372213.83X + 805.16, r = 0.9999; 1245713.26X + 4412.89, r = 0.9999; rosmarinic acid Y = 1713766.30X + 1387.01, r = 0.9999; salvianolic acid BY = 1209485.54X + 29336.62, r = 0.9998; RSD 1%), protocatechuic acid 98.5% (RSD 0.9%), lithospermate 98.3% (RSD 1.6%), rosmarinic acid 103.8% (RSD 0.7%), salvianolic acid B 98.3% ). Conclusion: The method is simple and easy to operate with good repeatability and good separation effect, which can be used as the quality control of Kangr heart capsules.
