目的:确定左西替利嗪对组胺诱导的单核细胞衍生的树突状细胞(MoDC)表达CD86和分泌IL-8的影响。方法:分离健康志愿者外周血单核细胞,进行体外诱导培养获得MoDC,以组胺作进一步诱导,并加入左西替利嗪进行干预,以FACS和ELSIA检测MoDC中CD86表达和IL-8分泌。结果:经组胺诱导后,MoDC的CD86表达水平和IL-8分泌量明显增多,左西替利嗪干预后CD86表达和IL~8分泌均受到不同程度的抑制(P<0.05)。此外,不同个体来源的MoDC对药物的敏感性也存在差异。结论:左西替利嗪的抗组胺作用机制是抑制组胺诱导的MoDC表达CD86和分泌IL-8,但临床药物治疗应个体化。
OBJECTIVE: To determine the effect of levocetirizine on histamine-induced monocyte-derived dendritic cells (MoDC) expression of CD86 and secretion of IL-8. Methods: Peripheral blood mononuclear cells from healthy volunteers were isolated and cultured in vitro to obtain MoDC. Histamine was used for further induction and added with levocetirizine for detecting CD86 expression and IL-8 secretion in MoDC by FACS and ELSIA . Results: After induced by histamine, the expression of CD86 and the secretion of IL-8 in MoDC increased significantly. The expression of CD86 and the secretion of IL-8 were inhibited by levocetirizine in different degrees (P <0.05). In addition, there are differences in the drug sensitivity of MoDC from different individuals. CONCLUSION: The antihistamine mechanism of levocetirizine is to inhibit histamine-induced expression of CD86 and IL-8 in MoDC, but the clinical drug therapy should be individualized.
