应用毛细管电泳前沿分析法研究了荧光素钠与牛血清白蛋白(BSA)之间的相互作用,利用游离荧光素钠平台峰高度和荧光素钠浓度与峰高的线性关系得出游离以及与BSA结合的荧光素钠浓度,将不同浓度比的荧光素钠-BSA相互作用体系进行电泳,考察结合情况,进一步采用非线性方程和线性方程对数据进行拟合处理,得到了荧光素钠和蛋白质在286和294 K下的结合常数及结合位点数;通过考察热力学参数推断二者之间主要以疏水作用力结合;借助计算机软件对二者之间的相互作用进行了模拟分析。
The interaction between sodium fluorescein and bovine serum albumin (BSA) was studied by the capillary electrophoresis frontier analysis. The linear relationship between peak height of free fluorescein sodium and sodium fluorescein concentration and peak height was obtained. The concentration of sodium fluorescein combined with different concentrations of sodium fluorescein-BSA interaction system electrophoresis, to investigate the combination of further use of non-linear equations and linear equations of the data fitting treatment obtained sodium fluorescein and protein in 286 and 294 K under the binding constants and the number of binding sites; by investigating the thermodynamic parameters deduced that the two mainly by hydrophobic interaction force; with the help of computer software to simulate the interaction between the two.
