目的研究人肽基脯氨酰顺反异构酶(简称 Pin1)、β-连接素(β-catenin)和细胞周期素D1(cyclin D1)在涎腺腺样囊性癌(SACC)中的表达,探讨其在 SACC 增殖和转移中的作用。方法采用免疫组织化学方法检测65例 SACC 中 Pin1、β-catenin 和 cyclin D1的表达情况,同时采用 Westernblot 和 RT-PCR 方法分别在 SACC83、SACC-LM、ACC-2和 ACC-M 4种 SACC 细胞株中检测 Pinl 蛋白水平和 mRNA 水平的表达。结果 65例 SACC 中 Pinl 高表达的51例(78%),cyclin D1高表达的41例(63%),二者的表达呈正相关(P=0.02)。14例(22%)SACC 出现β-catenin 在胞质中累积,其中6例(9%)表现为明显的β-catenin 的细胞核表达;其余51例 SACC 中β-catenin 仅在细胞膜上表达,伴淋巴结转移或浸润的病例中大都出现了β-catenin 在细胞膜上表达减弱或缺失(11/14)。Westernblot 和 RT-PCR 检测结果显示,Pinl 蛋白和 mRNA 在4种 SACC 细胞株中都呈现高表达。结论研究结果提示 Pinl 的过度表达在 SACC 中常见,其过度表达和β-catenin 信号传导通路的改变可能与SACC 的细胞增殖及癌的转移相关。
Objective To study the expression of Pin1, β-catenin and cyclin D1 in salivary adenoid cystic carcinoma (SACC) , To explore its role in SACC proliferation and metastasis. Methods The expressions of Pin1, β-catenin and cyclin D1 were detected by immunohistochemistry in 65 cases of SACC. Western Blot and RT-PCR were performed on SACC83, SACC-LM, ACC-2 and ACC- The Pinl protein and mRNA levels were measured in the strain. Results There were 51 cases (78%) with high Pinl and 41 cases (63%) with high expression of cyclin D1 in 65 cases of SACC. There was a positive correlation between them (P = 0.02). In 14 cases (22%), β-catenin accumulated in cytoplasm appeared in SACC, of which 6 (9%) showed obvious nuclear expression of β-catenin; β-catenin was expressed only in the other 51 cases of SACC Most of the cases with lymph node metastasis or infiltration showed the expression of β-catenin was weakened or deleted in the cell membrane (11/14). Western blot and RT-PCR results showed that Pinl protein and mRNA were highly expressed in all four SACC cell lines. Conclusion The results suggest that Pinl overexpression is common in SACC. The overexpression of Pin1 and the alteration of β-catenin signaling pathways may be related to cell proliferation and metastasis of SACC.
