目的:观察中肺合剂对小鼠Lewis肺癌肿瘤组织survivin、caspase-3基因表达的影响。方法:取C57BL/6小鼠50只,接种Lewis肺癌瘤株后,随机分阴性对照组(0.9%生理盐水),中肺合剂低、中、高剂量组(12.7g·kg-1、25.4g·kg-1、50.8g·kg-1),阳性对照组(环磷酰胺组,30mg·kg-1),每组10只。观察12天停药。无菌条件下取肿瘤组织,采用EnVision免疫组化方法测定中肺合剂对小鼠Lewis肺癌组织survivin、caspase-3蛋白表达的影响。结果:中肺合剂低、中、高给药组能不同程度下调Lewis肺癌小鼠肿瘤组织survivin蛋白的表达并上调caspase-3蛋白的表达,且均以中剂量组最显著,与生理盐水组相比统计学上有非常显著差异(P<0.01)。结论:中肺合剂可通过下调survivin表达和上调caspase-3表达,从而促进肿瘤细胞凋亡,抑制肿瘤组织生长。
OBJECTIVE: To observe the effect of Zhongfei Mixture on the expression of survivin and caspase-3 genes in Lewis lung cancer in mice. METHODS: Fifty C57BL/6 mice were obtained and inoculated with Lewis lung cancer tumors. They were randomly divided into negative control group (0.9% physiological saline) and low-, medium-, and high-dose groups of Zhongfei Mixture (12.7g·kg-1, 25.4g). ·kg-1, 50.8 g·kg-1), positive control group (cyclophosphamide group, 30 mg·kg-1), 10 mice in each group. Observed for 12 days to stop the drug. Under aseptic conditions, tumor tissues were taken and EnVision immunohistochemistry was used to determine the effect of Zhongfei Mixture on the expression of survivin and caspase-3 protein in Lewis lung cancer. RESULTS: The low, middle and high doses of Zhongbi Mixture could down-regulate the expression of survivin protein and up-regulate the expression of caspase-3 in Lewis lung cancer mice, and all of them were the most significant in the middle-dose group, compared with the saline group. Statistically significant differences (P<0.01). Conclusion: Zhongfei Mixture can down-regulate the expression of survivin and up-regulate the expression of caspase-3, which can promote the apoptosis of tumor cells and inhibit the growth of tumor tissues.
