目的采用新型中空纤维离心超滤联合HPLC测定人血浆中双氯西林的游离浓度。方法采用中空纤维离心超滤法对血浆样品进行预处理。采用Kromasil C18色谱柱进行HPLC测定,流动相为乙腈-25 mmol·L~(-1)磷酸盐缓冲溶液(p H4.5)=40∶60,流速1.0 m L·min-1,检测波长225 nm,柱温20℃。结果双氯西林在64.0~4 096.0 ng·m L~(-1)内线性关系良好(r=0.999 9),定量限为32.0 ng·m L~(-1),回收率为98.8%~100.3%,RSD<1.3%。结论本方法简便、快速、专属性强、灵敏度高,为双氯西林的游离浓度测定提供了一种新的、可靠的分析手段。
OBJECTIVE To determine the free concentration of dicloxacillin in human plasma by a novel hollow fiber centrifugal ultrafiltration combined with HPLC. Methods The plasma samples were pretreated with hollow fiber centrifugal ultrafiltration. The mobile phase was acetonitrile-25 mmol·L -1 phosphate buffer solution (p H4.5) = 40:60, the flow rate was 1.0 m L · min-1 and the detection wavelength was 225 by using Kromasil C18 column. nm, column temperature 20 ℃. Results Dicloxacillin showed a good linearity (r = 0.999 9) in 64.0-4 096.0 ng · m L -1 with a limit of quantification of 32.0 ng · m L -1 and a recovery of 98.8% -100.3 %, RSD <1.3%. Conclusion The method is simple, rapid, specific and sensitive. It provides a new and reliable analytical method for the determination of free chlorine concentration of dicloxacillin.
